See the basic poly details in the general questions. Sam’s mod is coded to be poly compatible, however, some dialogue and events are disabled for poly players due to the poly mod changes to the base game breaking these events.
Yes, I worked with one of the authors of DSV to add sprite and dialogue compatibility for Sam. Playing the trans version of DSV Sam does switch a few events in the spicy expansion to the core version for obvious reasons.

Not entirely, Sam has a confession and proposal event, the description and various other locations warn against confessing or proposing to him early. The mod will continue, but any events you didn’t see from the previous relationship status will be skipped. Some of these you can replay with the MH Event List mod.
The “No Evil” event arc blocks the sleepover series since Sam and farmer need to work out a misunderstanding. The events include Hear No Evil, Speak No Evil, See No Evil, and Homemade. You MUST see See No Evil and make a specific choice, OR see Homemade to re-trigger the sleepover series. If you started the series, then skipped to marriage, you will need to use MH Event List mod (or debug ebi in the SMAPI console) to force-play Homemade to trigger the Good Morning repeatable series. Technically yes, the dialogue you see will be from the mod that Content Patcher loaded last. However, if you care about the immersion/personality aspect of Sam, most other mods are not compatible, especially those with yandere or possessive storylines. Any mod that basically sticks with the vanilla portrayal of Sam should be story compatible.
Yes, you can play any expansion mod with Immersive Sam.
Most of them. Immersive Sam adds a small closet extension to his bedroom for a few events, so mods that change the interior of Sam’s room or add a room below his or between his and Vincent’s rooms (like the Bathrooms mod) are not compatible. Most recolor mods will work fine and completely blend with the closet addition.
